Internal energy(E) of a substance (atoms or molecules) is the sum total of the energy due to the molecular contribution of the substance, is called internal potential energy while contribution of energy due to motion of the molecules (translational, vibrational, rotational, etc.) is called internal kinetic energy. It is impossible to determine the absolute value of internal energy of a substance. Internal energy is a state property.
