Why is the gram stain so useful?  What is the principle of gram stainin?

  • The Gram stain procedure enables bacteria to retain color of the stains, based on the differences in the chemical and physical properties of the cell wall. 
  • Gram positive bacteria Stain dark purple due to retaining the primary dye called Crystal Violet in the cell wall.
  • The Gram stain is the most important staining procedure in microbiology. 
  • It is used to differentiate between gram positive organisms and gram negative organisms.
  • It is thought that this happens because the cell walls of gram positive organisms include a thick layer of protein-sugar complexes called peptidoglycans.

The principle of gram stainin

  • Gram staining is based on the ability of bacteria cell wall to retaining the crystal violet dye during solvent treatment. 
  • The cell walls for Gram-positive microorganisms have a higher peptidoglycan and lower lipid content than gram-negative bacteria. 
  • Bacteria cell walls are stained by the crystal violet.
